What are single amino acids?

Single amino acids are dietary supplements containing just one individual amino acid – such as pure L-glutamine, arginine, or beta-alanine.

Unlike complex amino acids, which blend several types together, here you know exactly what you are taking and in what amounts – which is ideal when targeting a specific physiological effect.

Every amino acid has a different job in the body. That is why single amino acids are selected based on your specific goal.

A quick note on terminology: Some substances in this category are not strictly amino acids in a chemical sense.

  • Beta-alanine is a non-proteinogenic beta-amino acid – it does not occur in proteins.
  • Taurine is an aminosulfonic acid.
  • L-carnitine is synthesised by the body from lysine and methionine.
  • HMB is a metabolite of leucine.

In sports nutrition, they are grouped here because they share the same origin and application: they act as targeted active compounds, not as building blocks for muscle tissue.

Single or complex? It’s about the task, not the quality

Simply put: complex amino acids are bricks, while single amino acids are the tools. You need both, but for different jobs. The question isn't which is better – it’s what are you trying to achieve?

Complex amino acids provide building materials

For a muscle cell to build new protein, it requires all nine essential amino acids simultaneously. While leucine acts as a trigger (the mTOR signaling pathway), protein synthesis will remain limited without the rest of the spectrum – it’s like starting construction without materials (Church et al., 2020).

This role is reliably filled by EAA amino acids or high-quality protein, typically in a dose of 10–20 g of EAA or 20–40 g of protein per serving.

Why not BCAA? They only contain three of the nine essential amino acids. According to Wolfe's review, they cannot trigger protein synthesis on their own – the body must take the missing six from the breakdown of its own muscle proteins (Wolfe, 2017). This doesn't mean they are useless, but they function as a supplement to sufficient protein intake, not as a replacement.

Single amino acids play a functional role

Individual amino acids are generally not building materials. They are tools – each for one specific mechanism:

  • Acid buffering. During a hard set, acidity accumulates in the muscle, and it starts to "burn". Carnosine acts like a sponge, soaking up those particles.
  • Nitric oxide precursor. Nitric oxide dilates blood vessels – hence that pump feeling.
  • Fat transport. Fatty acids cannot get into the cell's "power plant" (the mitochondria) on their own. They need a carrier.
  • Precursor for neurotransmitters. Tyrosine creates dopamine and norepinephrine – the chemistry of focus and motivation.

And here is the detail that changes everything: each of these mechanisms has a threshold dose. Below that level, the effect will not occur.

Now compare that to a typical broad-spectrum amino acid blend. It divides about 10 g across fifteen to twenty amino acids – meaning each one provides only a few hundred milligrams. That is far below the threshold where research shows an effect. Beta-alanine in such a mix won't increase muscle carnosine, tyrosine won't affect neurotransmitters, and carnitine won't reach doses used in studies.

That is precisely why single amino acids are sold separately. It isn't a sales trick – their effect simply stands or falls with the dosage, which cannot be achieved in a blend without the serving size reaching tens of grams.

Single amino acids by goal

Blood flow, pump, and vascular support

  • Arginine – a semi-essential amino acid and the primary precursor to nitric oxide, which dilates blood vessels. It has the strongest data for blood pressure: clinical study reviews report a drop of approximately 5.4/2.7 mmHg (Khalaf et al., 2019). Around training, it is used for blood flow support – however, study results are mixed, and the methodology of the main meta-analysis has since been professionally questioned (Viribay et al., 2020; Trexler, 2025).
  • Citrulline malate – bypasses breakdown in the gut and liver, effectively doubling blood arginine levels compared to oral arginine (Khalaf et al., 2019). Popular in pre-workout mixes. Watch the label: 8 g of citrulline malate ≈ 5 g of pure citrulline, the rest is malic acid.

Endurance and fatigue resistance

  • Beta-alanine – increases muscle carnosine, which dampens the rise in acidity during exertion. According to the ISSN position stand, it is ergogenic primarily for performance lasting 1–4 minutes – i.e., hard sets, sprints, intervals. It works by saturating stores, not from a single dose: it requires 4–6 g daily for at least 2–4 weeks (Trexler et al., 2015).
  • Taurine – an aminosulfonic acid involved in osmoregulation and antioxidant protection of cells. A meta-analysis of ten studies found a moderate improvement in endurance performance – the effect is real but small (Waldron et al., 2018). Also known as an ingredient in energy drinks.

Recovery and muscle protection

  • Glutamine – the most abundant free amino acid in plasma and muscle, and the main fuel for intestinal lining cells and the immune system. Let's be honest: the mechanism is well-described, but in healthy, well-nourished athletes, controlled studies have not yet proven benefits for immunity or performance (Gleeson, 2008; Ramezani Ahmadi et al., 2019). It is more interesting during periods of high physiological stress than as a daily routine.
  • HMB – a metabolite of leucine that dampens the breakdown of muscle proteins. Its best-documented use is in recovery, protecting muscle mass during dieting, and during periods of inactivity or rehabilitation – i.e., situations where the goal is to maintain what you have (Rathmacher et al., 2025).

Energy metabolism

  • L-carnitine – transports long-chain fatty acids into mitochondria where they are burned. It is studied in relation to energy metabolism and recovery. Frankly: EFSA has not approved any health claims for L-carnitine regarding weight loss or fat burning (EFSA, 2018). We describe its benefits as study findings, not as guaranteed effects.

Psychology, focus, and stress management

  • Tyrosine – the precursor for dopamine and norepinephrine production. Review work suggests benefits mainly when the brain is under pressure: acute stress, sleep deprivation, high cognitive load. Not generally for "better concentration" (Jongkees et al., 2015).

Other amino acids

  • Other amino acids – standalone amino acids outside the groups mentioned above, such as lysine, glycine, or methionine. If you are looking for a specific substance not found elsewhere, start here.

How to choose and combine single amino acids

You don't need to study biochemistry. Just answer one question: what do you want to solve?

  • Building or protecting muscle mass? Reach for complex amino acids – EAA or protein. You need a complete spectrum, not one substance.
  • Addressing one specific mechanism – pump, buffering, focus, fat transport? Choose a single amino acid in an effective dose. A blend won't cover it.
  • Want both? They aren't mutually exclusive. It is common to combine protein or EAA as a base with one or two targeted amino acids depending on your training goal.

Proven pairings: arginine with citrulline for blood flow, beta-alanine with creatine for repeated intense exertion – the ISSN notes a potential advantage for this combination, but only with an adequate dose and length of use (Trexler et al., 2015).

Safety and what to look out for

In recommended doses, single amino acids are generally well-tolerated. However, a few things are worth noting – we'd rather mention them upfront:

  • Beta-alanine: tingling is not a defect. The only commonly reported side effect is paraesthesia – tingling of the skin on the face, neck, or hands, which subsides within half an hour. You can mitigate it by splitting it into smaller doses (up to 1.6 g at once) or using a slow-release form (Trexler et al., 2015).
  • Arginine and citrulline may slightly lower blood pressure. If you are taking antihypertensives, consult your doctor about the combination.
  • Tyrosine should not be taken concurrently with MAO inhibitors and be careful if you have thyroid issues.
  • Glutamine is well-tolerated by healthy adults in acute doses of around 20–30 g (Gleeson, 2008). However, if you have liver or kidney disease, check with your doctor.
  • Monitor total daily intake. Beta-alanine, taurine, and carnitine are often found in pre-workout blends. It is easy to accidentally take them from two sources simultaneously.

Amino acids are not medicine and do not cure any disease. They are tools for fine-tuning – the foundation remains diet, sleep, and training.

How to select a single amino acid? Summary

Single amino acids make sense when you know exactly what you want to improve and want it in a dose that works according to studies. Pump and pressure → arginine or citrulline malate. Why does my skin tingle after beta-alanine?

It is paraesthesia, a harmless and well-documented phenomenon caused by how beta-alanine stimulates nerve endings in the skin. It appears within 15–20 minutes of ingestion and subsides on its own. You can mitigate it by splitting your daily dose into portions of 1.6 grams or less, or by using a slow-release form. It is not an allergy or a sign of an overdose.
